A STUDY OF THE RELATIONSHIP BETWEEN P53 MUTATION AND SMOHING
IN HUMAN NON-SMALL CELL LUNG CANCER

A study was undertaken during 1992 to 1993 to explore the relationship between p53 mutation
and cigarette smoking in human non-small cell lung cancer (NSCLC). The group of 43 males and 9
females, with age ranging from 30 to 71 years (median, 55), consisted of 30 squamous cell carcinomas
and 22 adenocarcinomas.
The detection of p53 mutations of all primary resected NSCLC specimens was performed using
inununohistochemistry. One patient with adenocarcinoma had a mutated allele of p53. PCR-SSCP
analysis showed the p53 mutation to be in the region between exons 5, 6, 7 and 8. Sequence
determination revealed that codon-280 had been changed from AGA to ACA.
Thirty-seven of the 52 lung cancer patients were self-reported smokers, suggesting that the
presence of lung cancer was probably associated with smoking. Among the 25 patients with a p53
mutation, the smokers accounted for (74%) 17 cases (68%), which was not significantly different from
the 20 smokers in the group of 27 showing no mutation in the p53 gene. These results implied that
the
p53 mutation may not be closely related with smoking cigarettes.
