Objective:
To study the possible relationship between pancreatic cancer
and previous chemical exposure and factors such as smoking
and a! ,hol consumption.

A case-referent study with two referent groups haa beea
designed to assess the association between chemical exposure
and pancreatic cancer. Historiea of chemical exposure related
to work, housing, diet, smoking, and other factors will be
obtaiaed from 100 incident cases of pancreatic cancer, 100
clinical controls frnm the same hospitals, and noocliaical
controls from the area population.
Results to Date:
The study is in a pielmunary phase.

Objective:
To study the relative mutagenicity of selected tobacco smoke
constituents using sister chr+omatid' eachanges (SCE) and the
Ames test.
Methods or ApPro.cl:
The induction of SCE in cultured human lymphocytes by
individual smoke compounds is studied using conventional
banding and fluorescence techniques.
Results to Dateo
Three compounds in a series of substituted methylindoles (2,3-
dimethylindole, 2,3,1-trimetbylindok, and' 2,3,b-tsimethylin-
dok) which have previously been shown to be mutagenic by
the Ames test have been found to eauae SCE.

Objective:
To (1) analyze the actions of acvte and chronic nicotine
treatment and exposure to cigarette smoke on the various
hypothalamic catecholamine nerve terminal Bystems of rats in
various endocrine states; k2) relate the actions on these systems
to effects on the secretion of luteiniring hormone (LH), follicle-
stimulating hormone (FSH), prolactin, adnaooortimtropic
hormone (ACTH), corticosterone, growth hormone, thyroid-
stimulati.n8 hormone (TSH), and vaaopresaia;,and (3) study, by
means of quantitative immunocytocDemistry, the possible
direct effects of chronic niootine and exposure to cigarette
smoke on LFIItJi, somatoatatia, and' TRH immunoreactive
nerve terminals of the median eminence to discover possible
signs of aetions on peptidergic cells.
Metloda or Appeoaeh:
Biochemically regional catecholamine and SHT stores are
analyzed by amine fluorescence histochemistry, quantitative
immunocytachemistry, aod hi8'b pressure liquid chnomatogn-
phy with electrochemical detection. and radioimmuao.asays of
hypophyaeaW hormones are made.

The results show that acute exposure to cigarette smoke or
niootine injections produce widespread activations of hypotha-
lamic dopamiae and noradrenaline n.erve terminal systems
which probably are produced' via activation of nicounic
cholinergic reoepton controlling the synthesis in the release of
catecholamina in the various parts of the hypothalamus.
Furthermore, nicotine in repeated doses prod'ucea lowering of
LH, FSH, and prolactin secretion. effects which seem to be at
least in part mediatad via activation of the tubervinfundibular
dopamine neuronss. Nicotine can produce reductions of discrete
hypothalamic noradrmalinc levels within a few minutes and
can also markedly eahanee the secretion of ACTH and
vaaopra.rin, actions which also may involve effects on hypo-
thaLmic noradrenergic mechanisms in discrete areas.

Studies on chronic exposure to cigarette smoke are planned so
that the effects of smoking ia a chronic situation can be
compared with chronic exposure to niootine.

A test systeatm, baaed on the ability of tumor promoters to
induce viral antigen syntbesis in Epatein Barr Virus carrying
Burkitt lympboma cells when combined with n-butyrate, is
being uaed The presence of a synergistic eNect between n-
butyrste and tumor-promoting phorbol eatas, resulting in an
early antigen induction, has been shown to correlate extremely
well with the tumor-promoting activity of the rsspective
phorbol eater in miee. Fractions of tob.ooo smoke are currently
being tested for tumor promoters.
