Bethesda, MD 20816 Dear Dr. Henry: We have the following comments concerning Study No. 1-2486.003.001, "Comparative Toxicity of Test Cigarette Smoke and Reference II Cigarette Smoke with Reference I Cigarette Smoke After Inhalation in Mice (D29 and D30)," Draft Report. 1. On the title page, line 3 of the title: "After" should be capitalized. 2. On page 4, line 3: "cigarette" should be plural in the phrase "Reference II cigarettes." 3. On page 6, paragraph 3, line 2: "randomization" is miss- pelled. 4. On page 11, paragraph 3: a. Lines 6-7 - the text should read "survival to smoke exposure in all other groups was 100%." b. Line 9 - the text should read "there were no significant differences." - 5. On page 11, paragraph 4, line 12: the text should add that at RSD2, mice either gained weight or remained at the same weight during the 24 hours after exposure. 6. On page 13, the conclusion recommends that the dosing regimen for repeated dose studies be set at 7 puffs per exposure. This needs some sort of explanation, because both RSD1 and RSD2 were run in this study at 8 puffs per exposure. Survival was 100%, and all animals received the scheduled number of doses. Such an explanation appears in the Draft Report 1-2130.003 with D13 and D17, on page 14 (copy en- closed).
