Henry ~1e LOCATION: SUBJECT: Equipment Purchase We would like to formally request the use of existing contract funds for the purchase of three pieces of equipment necessary to our ongoing and proposed research. As we have discussed with you over the last few months, the experimental approaches described in our progress reports and proposed studies require the capability of a high-pressure liquid chromatography (HPLC) system. Such a system would allow sensitive evaluation of and identification of metabolites and proximate carcinogens in both our in vitro and in vivo studies. The versatility of the HPLC woulU-a ~tlonally aTTow its use as an adjunct technique in the DNA damage/alkaline elution assay and dosimetry studies. Almost $10,000 was budgeted on Contract 0014 for this equipment, however, approx:mately $10,000 more will be required for purchase of a system which will fulfill our needs. We have arranged to purchase a demonstrator model at substantial savings (see attached page). The second major piece of equipment is a set of polycarbonate exposure modules for the SEM. Originally each SEM was to be pro- vided with two sets of eight modules so that one would be in use while the other was being cleaned. Because of the substantial cost of this system, we wished to fully evaluate it before requestingg one additional set. No major redesi gns of the modules are envisloned. Due to our budgetary constraints in 197t3-79, and the resulting decrease in personnel (9 lab aides In 1977-78, 6 lab aides.in 1978-79), an additional set of modules Is necessary so that the same number of mice will be exposed to smoke next year as during this last contract year. The reason for this is the tiine saved by having one set In use while the other is being cleaned. We must point out that we are requestln? a set of modules for only one of the two SEMs that are in operation here. The third piece of equipment is an Aminco-Bowman spectrophoto- fluorometer. As we have discussed with you, the spectrophotofluoro- meter available for use on the CTR contracts was funded from other agencies and continued use and accessability for future work is doubtful. A description of the items follows. Unless jou object, we will order these items so as to take advantage of the availability of the demonstrator HPLC.
