This application had been active for only 5 months at the tims that the progress report was submitted. k-methylumbeliferone (4-M.i) was used as an exogenous substrate for g1ucUronidatibn by a line of the RP.~ of cultured human lymphocytes. The reaction could be prevented by beta-glucuro- nidase or saccharolactone. They are now setting up assays with other exogenous substrates and developing analytical procedures for B-a-P conjugates. The glur_uronyltransferase (GT) could be removed frcm. lymphocytes by a combination of freeze-thaw and homogenization. It was stable at -70* for at least a week. GT was iipduced by 5ugDBA; 7x for AHH, 2x for 4-.D4T, and 1.5x for testosterone. Four different lines of lymphocytes have been studied and at least a lOx difference in GT activity has been noted. Mure #.ines are being run using the testosterone GT reaction. Projection. Unchahged from last year when submitted.
