Rasco, Ph.D., Assistant Professor of Biology, The University of Texas System Cancer Center, M.D. Anderson HospiL:al and Tumor Institute, Houston. "Hydrocarbon Metabolizing Enzymes and Lung Cancer." Although Dr. Rasco is ltsted as principal investigator, Dr. Toshio Yam&uchi is also project investigator. The latter writes the procedural outline and was trained in immunology, the former in biochemistry. They both have been working for about two years on the development and checking of methods of AHH determination and their studies to date are reviewed in the preprints and manuscripts submitted with the application. They have ttade three major changes in method and report their results in three parts. The percent of exclusions has decreaced of data on individuals with each procedural modification. The patients with lung cancer have generally had, as a group, higher AHH values than non-cancer or other cancers with the exception of squamous celled cancer of the oropharynx. The distribution of AHH levels tends to be unimodal and probably polygenic. Actually they have no good genetic data. Proposal. They plan to begin 3H-BaP method of assay. This will determine alll water soluble metabolites of the PAN used for inducer (not just 30H-BaP). This method is now used at Creighton. They will carry eight donors at periodic intervals to serve as standards and set up an exchange with Creighton and MA that will exchange similar samples that will all be run on one time in the three laboratories and with similar materials. Second, they will continue the readings on lung and oropharyngeal cancers to inereaEe the numbers of observations. Samples from these populations will be "i.ndex" numbers for a proposed family study of 30 in each group, controls and the two types of cancer. Age is important. The significant data were on persons under 40 years. There was no difference between smokers and nonsmokers, stopped, or never smoked. The budget is for one technician and expendable supplies and for the first year includes a C02 incubator, approximately $2,000. Dr. Shaw has two other grants totaling about $96,000: one for AHH and colon cancer, and one titled "Microsomal Enzyme Studies." The budget seems reasonable. I W.U.G. WUG:ek
