Los Angeles County Harbor General Hospital, Torrance, CA, • 90502 New Application No. 1128 "Effect of Cigarette Smoking on Cytochrome P-448-Associated Enzymes." The applicant is Associate Professor of Medicine and Chief of the Section of Hepatology at the Harbor General Hospital, Torrance, Ca. His preparatory and medical training were in England. Since 196e he has been in the USA, first as a research fellow and since 1976 at his current aCdress. He has done considerable recent work with Vesell and with amino- pyrene metabolism using breath analysis methods. He has a grant of approximately $60,000.00 from N.I.H. for work on liver function and has another application pending. The greater part of the request is for professional and tech- nical salaries. In the first year $33,000.00 is requested for a high pressure liquid chromograph and a spectropho;,ometer. Volunteers will be used, 20 with lung cancer and smokers, 20 without lung cancer and smokers and 20 nonsmokers. Aminopyrene (P-450), theophylin and phenacetin (P-448) and G-S-T will be followed in all patients. Aminopyrene will be determined by 1I+C determinations in breath after injection of labelled compound. Theophyline will be determined in breath 11+C02, and in blood, the need for the high pressure liquid chromatograph, and B.S.P. will be used for the glutathione-S- transferase. AHH will be determined by determining 30H-BaP formed in monocytes in vitrc:. In addition, 600 neonates will have blood samples drawn 2 and 8 days after birth, de-`z iled histories of the mothers will be taken concerning smoking and drug habits and the bilirubin foreiaticn and metabolism in the nawborn will be determined. The investigator seems competent. I am not acquainti:d with the research environment of the Hospital. I suggest that Drs. Sommers and Jacobson attempt to visit the applicant when on the way to or from La Jolla on the Fishman visit. VFL might try to set this up. WUG/mb
