The principal investigator and co-principal investiaator have hac3 previous research support from GTR. It is not easy to deterrmine just what sane of them will be doing, particularly in the Wisconsin group with background training in Sweden, Ireland and Australia, as well ds Iowa ancl Wisconsin. It is also difficult to determine }ww much has been acroirplishod durinq the past year because so of the titles are of abstracts of papers presented at meetings or of reviews in monographs. The application of RIA and immunocyto- chemistry has faci7.itatec# studies of a large nwrber of neural peptidPs, some first observed by Von Euler 50 years ago. Drs. Polak and Bloom have certainly been recent leaders in this field. The localization of a number of groups of polypeptides ranging from 5 to 28 amino acids in brain, gut, lung, skin, etc., an.' their association with unfolding neuroendocrine functions and diseases is increasing rapidly'. 'I'heir association w-ith cardiovascular and respiratory functions put's them in an area of interest to CTR. A peptidergic autone-ic systam mist now be added to the autc>ncmic adrenergic and cholinerc;ic sr•staiu . (It is' interesting that Von,Euler also discovered pQpe.ridine in the brain in the 30's.) Proposal. T9iey propose the collection of tissues from nor.i.en°s with puilimnary di.seases, particularly asthma and chronic bronchitis from 100 cases per year for estimation of neuroendocrine (NE) cells. Wisconsin will aJ.so provide sc,e material.. A njmber of regulatory peptide abnormalities may be found. At least more might be learned of the regulatozy peptide functions in health and d.isa3se. A Regulatory Peptide Unit is proposed for the London area. Protocols are proposed for the estimation and the localization of neuropeptides and bioaenic aminesr for the ultrastructural localization in tissues and cells, and for their response in sensitized ani.mals. 5ome physiological studies will be done on thc caro.id body receptors and,oxygen levell mbnipulation. Will plans to infuse pqptides into hearts, measure peptide release in blood and lymph, and measare coronary pressure in collaboration with a Professor Sc.'iaper at Bad Nauheim, West Germany (another internav.or.al oollaborator) . Budget. Zhe Wisconsin Laboratory needs a freeze 8r±, liphilizer and moneA• for 20% of a mass spectroarapah ($23,000) and the London Laborator?• a refrigerated centrifuge and a Polyvar Namirsky Microscope ($40,000). Travel expeazses are also high. The high radionucides are really expensive and needed. Cmnent. 7hese shctiuld be separated. The Wisconsin unit has contributec: relatively ls~ttle to date and that related to effects of hypobaric and hyoerbaric states. I am not impressedwith the program at Wisconsin.
