Becker, M.D., and Gregory Siskihd, M.D., Cornell University Medical College Application No. 1010 "Investigation of the Role of Allergy to Tobacco Constituents in the Pathogenesis of Arteriosclerosis anet Myocardial Infarction" Discussion: Since its first submission, this proposal has been somewhat refined and simplified. Tobacco leaf glycoprateiin (TGP) will be extracted, and a tobacco leaf lipid (TLL) also will be prep.3red. The TGP will be incorporated into Sepharose beads as an insoluble absorbant, and this material will be tested with various immunoglobulin classes derived from serum. If antibodies belonging to the IgE class can be demonstrated, volunteers will be skin tested. Subjects to be studied will include medical students and smokers with heart disease and thromboangiitis obliterans, as well as a group derived from persons who have given up smoking. Absorption of TGP will be studied and its presence in serum and saliva will be detefrmined. Tobacco leaf lipid also will be tested for allergenic ectivity. The sites of serum injection in monkeys from subjects with high levels of IgE antibodies to TGP will be injected with the lipid extracts. Cri tigue : Ttiis proposal, while still fairly elaborate, is now reasonably consonant with the critique provided at the previous meeting of the CTR. The budget is appropriately modest.
