Gardner SUBJECT: Application 599-C. Domingo M. Aviado, M.D., University of Pennsylvania School of Medicine, Philadelphia. "Influence of Cigarette Smoke on Pulmonary Emphysema and Bronchospasm." (second year of a 2-year grant but he's asking for 3 years.) A research proposal of six parts is presented. Thirty monkeys, 150 rats and 300 mice will be smoked for six months (controls also in figures) and pulm,:•nary functional studies will be done. Also C0, skatole and nicotine will be given. Antitrypsin and blood gases will be followcd. Pathological studies uill be done. Since 1967 - 5 papers related to this project. !1e has also been doing much work on antimalariais. He has an F.D.A. grant on "Cardior.ulmonary Toxicity of Propellants for Aerosols" ($:4,000) and $37,000 from Departrient of tne Yrmy on "Drug Therapy of Acute Respiratory Insufficiency." ProAress , No blood vessels occurred in dogs, rats or hamsters exposed to cigarette smoke 4-10 weeks. Racs given smoke showed increase in antitrypsin but no emphysema. Papain produced emohysema. E'xperiment on :mnke on phospholipids underways generally a decrease. Smoking causes bronchosoasm, increased pulmonarf resistance and decreased tidal volume. Same strain differences. Paraquat (3 weed killer) reduces surfactant given :ntrareritoneally. It causes pulmonary conger•stion, increasc•s antitryp3in and decreases phospholirids. A nice quantitative report. W.U.i; . WUG:ek
